Depletion of NET inhibits Notch1 signaling in human colon cancer cells. HCT116 and SW480 cells were treated with NET-targeting siRNAs (siNET1 and siNET2) or negative control siRNA (siNC). After 48 h, cell lysates were harvested, and the protein samples were separated by SDS-PAGE. The levels of full length Notch1, cleaved Notch1 and <t>Snail1</t> were detected by western blotting, and GAPDH was used as the loading control. The band intensities of NET relative to GAPDH were quantified and normalized to the siNC sample. The data are representative of three independent experiments. One-way ANOVA was used to compare data between siNET- and siNC-transfected cells.
